报告摘要:As the rapid development of nanotechnology, nanomaterials have been proposed to serve as functional units for biomedical application in many innovative ways. Therefore, the detailed knowledge of interactions between engineered nanomaterials and biosystems has become increasingly important. Herein, I will start with my study of nanoparticles interacting with human cells, revealing their intracellular fate inside cells. By using advanced microscopy technologies, the endocytosis and exocytosis are identified as the major pathways for nanoparticles entering and exiting the cells, respectively. Most of the nanoparticles are found to be enclosed in membrane bounded organelles, which are stable as very few nanoparticles escape into the cytosol. I will then introduce my development of multifunctional composite nanocarrier systems for biomedical application, with unique selfdecomposable feature which aims at achieving safe excretion from the biological system and enhancing drug efficacy. By combing with the optical switch on the developed nanocarriers, I managed to control the spatiotemporal release of drugs in cytosol, circumventing multidrug resistance and reducing the systemic toxicity of the drug. The obtained understanding on above mentioned studies is critical for nanoparticlesbased drug delivery and intracellular sensing, can lead to a number of exciting applications in biology and medicine.
